Background

Kinases and phosphatases are responsible for phosphorylation and dephosphorylation, respectively. Phosphorylation is of particular importance in biology because it is a key reaction in protein and enzyme function, glycolysis, and energy storage and release. Many types of molecules can be phosphorylated. Protein phosphorylation, glucose phosphorylation, and oxidative phosphorylation are the most important types of phosphorylation. Phosphorylation plays a key regulatory role in both prokaryotic and eukaryotic cells. It has long been recognized as an essential mechanism for controlling cellular function. In particular, protein kinases are prominent in cellular signaling transduction. Their hyperactivity, malfunction or overexpression has been implicated in several diseases, such as cancer. Therefore, analysis of phosphorylation is important to elucidate its mechanisms. A wide range of methods and techniques such as electrophoresis, isotope labeling, immunoassays, and mass spectrometry have been developed to determine whether a molecule is phosphorylated and to further identify and characterize the phosphorylation sites.
