Background

Reversible protein phosphorylation is a common mechanism used by cells to propagate or terminate cell signaling. Kinases and phosphatases alter protein conformation and/or protein interactions by the addition or removal of phosphates. Phosphorylation/dephosphorylation reactions have an impact on substrate activity, half-life, function, and their subcellular localization. The antagonistic activities of these enzymes are involved in the regulation of multiple cellular processes. Therefore, the underlying molecular mechanisms controlling phosphorylation/dephosphorylation are of great physiological importance. In fact, phosphatases have been largely neglected while most kinases have been well characterized.

Imbalance between phosphorylation and dephosphorylation have been demonstrated to be implicated in the development and progression of a myriad of diseases, making kinases and phosphatases promising therapeutic targets. Due to many difficulties in studying phosphatases in the past and unsuccessful attempts to develop drugs targeting them, they have long been considered undruggable. However, tremendous progress has been made in phosphatase research over the past 20 years. New and creative methods for phosphatase screening and profiling have been developed to address specific challenges, paving the way for the development of drugs targeting phosphatases. Phosphatase have regained a strong attention.
