Kinase Substrate Screening

Creative BioMart offers a wide range of service options to help our customers identify potential substrates for various types of kinases. Our talented scientific team is able to provide our customers with unparalleled expertise and experience in kinase biology and pharmaceutical research.

Protein phosphorylation is an important post-translational modification (PTM) that mediates intracellular signaling in a variety of cellular processes. More than 500 kinases encoded by the human genome are responsible for the regulation of phosphorylation events and thus are involved in many physiological and pathophysiological processes. In general, a single kinase has multiple substrates/phosphorylation sites. Although our understanding of kinase biology has improved tremendously, the specific substrates of each kinase have not been fully elucidated and these phosphorylation sites remain largely unknown. Therefore, comprehensive kinase substrate screening is necessary to understand kinase signaling networks and kinase-substrate interactions involved in disease pathology, thus contributing to the development of more specific kinase-targeted drugs.

Recent advances in mass spectroscopy and proteomics methods have enabled rapid profiling of the phosphoproteome, allowing accurate identification of phosphorylation sites and comprehensive analysis of dynamic phosphorylation events within cells. However, the identification of kinase-substrate pairs remains an exceptionally challenging task. In recent years, a variety of approaches have been continuously developed to characterize the relationship between kinases and their direct substrates and to elucidate signaling pathways. These advances in kinase substrate screening technology have the potential to transform our ability to identify new kinase targets.

Service Options

Genetic screening

Analog-sensitive kinases with specific mutations within the ATP-binding pocket can be used in various ways to identify substrates. We provide chemical genetic screening by generating kinase mutants and ATP analogs to label the direct substrates.

Protein and peptide microarrays

Protein and peptide microarrays allow purified kinases to be incubated with each putative substrate in a high throughput fashion. We use customized protein and peptide microarrays to help our customers effectively detect substrate of various kinases.

Phage display-based substrate screening

Phage display provides a fast and powerful technique to identify candidate substrates by sequencing phage plaques containing positive phosphorylation signals. We provide phage display-based approaches to perform substrate screening and determine kinase substrate specificity.

Protein interaction-based substrate screening

Identification of kinase-interaction proteins is a common practice for screening potential kinase substrates. We offer a variety of methods for protein-protein interaction analysis including yeast-two-hybrid approach.

Mass spectrometry-based high throughput screening

Mass spectrometry (MS) play a dominant role in proteomics study. We have established a state-of-the-art MS platform for the identification of phosphoproteins and characterization of their phosphorylation sites.
