Background

Reversible phosphorylation and dephosphorylation are important regulatory mechanisms in both prokaryotes and eukaryotes. Kinases are responsible for catalyzing the transfer of phosphate groups from nucleoside triphosphates to acceptor groups. Their role in mediating important cellular processes and signal transduction pathways have been extensively studied. Deregulation of kinase activity has been associated with a variety of pathological conditions, such as cancer, cardiovascular diseases, and inflammation. As a result, kinases have emerged as attractive targets for drug discovery.

An effective method for monitoring kinase activity is essential for kinase research and drug development. Radiometric assays with the advantage of high sensitivity are the conventional analytical technique for kinase activity. However, these assays are time-consuming, expensive, and have a high risk of radioactive contamination. Alternative assays such as colorimetric assays and fluorescence-based assays have been developed to overcome these limitations, but they still have some drawbacks such as labor-intensive procedures, the need for labeling, and expensive instrumentation. Label-free methods have attracted increasing attention in the development of kinase assays due to their low production cost, pathway-unbiased, fast response, and high sensitivity. These assays provide a new dimension to elucidate kinase biology and facilitate drug discovery.
