Background

Protein phosphorylation is a widely studied post-translational modification (PTM) involved in almost all cellular processes, spanning from cell cycle to metabolism. It has been reported that phosphorylation sites display tissue-specific preferences. The phosphorylation of cellular proteins is tightly controlled by kinases and phosphatases with different expression levels in different tissues, thus phosphorylation and dephosphorylation are dynamic events with restricted spatial and temporal distribution. The activity of kinases and phosphatases are themselves fine-tuned by phosphorylation events, generating a complex regulatory pattern.

Indeed, deregulated signaling pathways is usually a hallmark of many diseases. Phosphorylation pathways have also been shown to be implicated in the pathogenesis of several severe diseases, such as cancer and diabetes. Kinase/phosphatase genes with elevated expression levels in particular tissues can serve as a starting point to understand their biological functions and the underlying mechanisms of related diseases. This highlights the importance of developing techniques that allow for comprehensive and quantitative determinations of tissue-specific phosphorylation.
