Background

An idiotype can be defined as a set of idiotopes present within the antibody complement determining regions (CDRs). Anti-idiotypic antibodies (anti-ID Abs) can bind to the idiotope of another antibody and are therefore antibody specific. These anti-idiotypic antibodies, capable of binding to therapeutic antibodies within biological fluids, are powerful tools for immunogenicity assays and PK/PD analysis during the development of therapeutic antibodies, allowing measurement of free or total drug levels in preclinical and clinical samples. In addition, they can also be used as positive controls in anti-drug antibodies (ADA) assays.

Protein phosphorylation, which plays a key role in many physiological processes, is often deregulated under pathological conditions. The current understanding of how kinases and phosphatases regulate phosphorylation events and thus coordinate cellular and organ functions makes them potential drug targets for the treatment of many diseases. Antibodies targeting kinases and phosphatases have been one of the most active and promising pipelines in the pharmaceutical industry. Anti-idiotype antibodies are expected to be used to better understand the efficacy of existing monoclonal therapeutics, to develop new therapeutic antibody candidates, and to evaluate biosimilars.
