Background

Protein phosphorylation is an important cellular regulatory mechanism. A broad range of receptors and enzymes can be activated/deactivated by phosphorylation/dephosphorylation events. Kinases are responsible for protein phosphorylation. They catalyze the transfer of phosphate from a donor ATP molecule to a wide variety of cellular substrates, resulting in the formation of phosphorylated products and ADP. Kinases themselves can also be regulated by other kinases and initiate cascade reactions. Because of their important role in many cellular processes, kinases have been one of the most widely studied enzyme families in biological and medical research. Hyperactive or malfunctioning kinases have also been associated with various diseases such as cancer, diabetes, and cardiovascular diseases, making them important targets for drug discovery.

The rapidly growing interest in kinases research and drug discovery has prompted the development of many different assay technologies to monitor changes in kinase activity. Kinase functional assays focus on detecting kinase activity by directly or indirectly quantifying either the formation of the phosphorylated product or the amount of ATP used or ADP formed. Compared to binding assays and cellular assays, kinase functional assays are the most preferred assay format. The commonly used functional assays include colorimetric assays, bioluminescent assays, fluorescence-based assays, radiometric assays, label-free assays, and mobility shift assays. Understanding different format of kinase functional assays is the first and key step in elucidating kinase signaling and developing successful drugs.
