Background

In both prokaryotes and eukaryotes, protein post-translational modifications by reversible phosphorylation with kinases and phosphatases regulate key signaling pathways in cells. The addition or removal of a phosphate group may result in altered protein activity, subcellular localization, or interactions. The relationships between substrates and kinases/phosphatases are vital in cellular homeostasis. The flow of information in cells can be directed via reversible protein phosphorylation. Kinase/phosphatase signaling pathways appear to be staggeringly complex, as many molecular details are still poorly understood.

Understanding the complexity of phosphorylation is of great significance. Aberrant phosphorylation has been demonstrated to cause or contribute to a wide variety of human diseases, making kinases and phosphatases promising therapeutic targets. Structural characterization of kinase/phosphatase family has made a significant contribution to our understanding of molecular mechanisms regulating their activity. The crystal structures of several kinases and phosphatases have been determined, providing more insights into underlying molecular mechanisms of cell signaling & regulation, and promoting innovation in drug development.
