Background

Kinases play a key role in the regulation of signal transduction and many other cellular processes. Most of these physiological processes are regulated by molecular mechanisms involving kinase-ligand interactions. Ligand molecules can induce conformational changes to regulate kinase activity. Since kinases have been intensively studied as potential therapeutic targets for various diseases, quantification of ligand binding to kinases by ligand binding assays is essential not only for understanding biological functions of kinases but also for developing new drugs based in large part on binding affinity and efficacy. Every step of kinase-ligand interactions can be analyzed by different ligand-binding assays. The data of in vitro ligand binding may also provide important information about in vivo efficacy of these ligands.
