Background

Kinases transfer the γ-phosphate of ATP to hydroxyl groups in specific substrates. The human kinome encoding 634 kinases (pseudokinases included) comprise one of the largest families of homologous proteins. Kinases play an integral role in cell signal transduction. Overexpression and/or dysregulation of kinases result in many diseases such as cancer, neural disorders, inflammatory diseases, and metabolism problems, making them one of the most promising groups of drug targets. Currently, approximately one quarter of all research expenditures on drug development are focused on kinases.

Given the clinical importance of kinase-targeting drugs, many technologies and platforms have been developed for identifying kinase substrates and discovering small molecules against kinases. Over the past few years, numerous biotech and pharmaceutical companies have established high-throughput platforms for kinase screening.
